土壤真菌提取物生物活性研究及LC/MS-QTOF代谢物分析

土壤作为微生物的重要生态位,是发现具有药学应用潜力的生物活性天然产物的重要来源。本研究旨在调查从沙特阿拉伯不同沙漠地点分离的三株真菌菌株的生物活性和代谢特征。从沙特阿拉伯的Al-Qasab、Tabuk和Almuzahimiyah地区采集土壤真菌菌株并进行鉴定。采用血琼脂、营养琼脂和沙氏葡萄糖琼脂培养基,检测菌株对金黄色葡萄球菌、粪肠球菌、肺炎克雷伯菌和大肠杆菌的抑菌活性。真菌提取物对人乳腺癌和结肠腺癌细胞的活性/增殖进行了评估。采用高效液相色谱法(HPLC-DAD)耦联LC-QTOF-MS分析方法对真菌乙酸乙酯粗提物进行代谢物分析。鉴定的三株真菌菌株为Chaetomium sp.、Bipolaris sp.和Fusarium venenatum,其对不同微生物的抑制活性随粗提物、菌株类型和培养基的不同而变化。F. venenatum提取物相比Chaetomium sp.和Bipolaris sp.提取物对四株癌细胞系表现出更强的抗癌活性。HPLC和LC/MS-QTOF筛选从Chaetomium sp.鉴定出9种化合物,从Bipolaris sp.鉴定出3种化合物,而F. venenatum提取物中的化合物未完全鉴定。研究结果表明真菌提取物的某些生物活性在体外得到验证,提示这些提取物可作为寻找具有医学应用前景的活性物质的起点。需要进一步研究以完全鉴定F. venenatum提取物中的生物活性分子。
